Research findings of - G - 7 CAPSULES


Studies and evaluation done on G - 7 Capsule presented here are proprietary formulation of Dr. JRK's Siddha Research and Pharmaceuticals Pvt Ltd



G-7 capsule, the proprietary formulation of Dr.JRK's Siddha Research and Pharmaceuticals Pvt.Ltd., Kuntrathur, Chennai was studied.

Both In vitro and In vivo study was carried out for confirmation. The peritoneal mast cells were used for the in vitro study. The isolated mast cells were maintained in RPMI 1640 tissue culture medium in a flat bottom multi welled micro titre plate. The cells were treated with different concentrations of G-7 extract prepared in Hans buffer with the aid of Myosol as solvent. The concentrations of the G-7 tested were 2,10,20,40 and 50 µg / ml of medium. The treatment time was kept constant for 20 minutes. Later, the cells were challenged with sensitized IgE and the opsonized allergen. The amount of histamines and neutral proteases released by the G-7 treated and untreated mast cells were quantified. Histamine quantification was done by radio immune assay using radio tagged antihistamines using Scintillation counter. Similarly, attempts were made to check the histamine release by using specific immune staining.

Proteolytic activity was determined by substrate-enzyme reaction assay. The end product the tyrosine released as a result of proteolyses of collagen is determined.

Determination of mechanism of action
The IgE – FC receptor site in the mast cells were blocked with a dummy protein prior to G-7 treatment. The dummy protein is later removed and was challenged with sensitized IgE. The findings revealed that the degranulation inhibition is mediated through IgE receptor binding by the G-7 moiety.

In vivo study

Dust inhalation study


For in vivo confirmation of the effect of G-7 in mast cell degranulation inhibition, dust inhalation study was conducted using Rabbit model. Rabbits were pre-treated with G-7 for 15 days. Later the treated and untreated animals were challenged with house dust through a nebulizer in a dust inhalation chamber. The histamine released in the system is assayed as mentioned above.

The findings of the study reveal that G-7 offers greater protective effect to the mast cells from degranulation during the allergen exposure

Inhibition of broncho spasm during allergen exposure and anaphylactic shock in the case of - G -7 treated animals also observed in a separate study.
